Biotinylation is the attachment of biotin to proteins or other biological macromolecules and can be used to target specific functional groups such as primary amines, sulfhydryls, carboxyl groups and carbohydrates. Biotin-avidin/streptavidin systems are often used for efficient detection or purification of proteins. Sulfhydryl groups, namely sulfhydryl groups (-SH), are also a commonly used class of biotin labeling sites. The thiol group exists on the surface in the form of a disulfide bond (sometimes in a free state), and after reduction, it selectively reacts with maleimide in the form of mercapto alcohol to form a stable thioether bond. The addition reaction has fast reaction and high selectivity under the condition of neutral pH, and is a commonly used biomarker reaction. Compared with amino labeling, the labeling site of thiol biotinylation is limited to cysteine, and the labeling has little effect on the biological activity of the protein.
