Single Targeted Drug Delivery System

Single Targeted Drug Delivery System

Cancer is one of the leading causes of death worldwide, and achieving highly effective treatments for it remains challenging. Currently, the effectiveness of cancer therapy depends largely on tumor-specific drug delivery. Currently, antibody-drug conjugates (ADCs), drug conjugates, and nanocarriers have been used for efficient targeted drug delivery to treat cancer. In order to specifically target and eradicate cancer cells, a precise distinction between tumor cells and normal cells is required, and future nanomedicine may require new design principles to achieve active targeting of nanocarriers. Small biomolecules such as biotin (BIO) exist in cells and bind to receptors with high affinity due to their small molecular weight and small size. Biotin receptors are upregulated in a variety of cancer cells, while being expressed at low levels in normal cells and tissues, making them useful as targeting ligands for cancer chemotherapy.

Biotin as targeting ligand

Biotin, also known as vitamin H or coenzyme R, is an essential cofactor for carboxylase activity. Biotin is involved in important metabolic processes related to gluconeogenesis, fatty acid synthesis or BCAA catabolism. Biotin promotes cell growth and can be biotinylated by binding to different molecules through the valeric acid tail. Biotin enhances target cell/tissue internalization of nanoscale drug delivery systems and is an active strategy for cancer-targeted therapy. The surface of the drug delivery system (DDS) can be functionalized with biotin by conjugating biotin to the polymer chain for efficient drug delivery.

Development and prospects

Currently, future nanomedicine based on drug delivery systems for anticancer drugs could utilize biotin for efficient targeted drug delivery systems. Biotin is commonly used as a targeting ligand for cancer therapy, enabling recognition of specific receptors in human cancer cells. In the future, biotin is expected to be used in a wider range of cancer treatments.
