Feature | Biotinylated antibodies typically maintain their biological activity because the biotin group is relatively small. Conventional dimerization methods, such as using amine-NHS esters or converting amines to thiols followed by maleimide chemistry, result in random biotin attachment on antibodies. In some cases, biotin labeling in specific Fab regions may interfere with antigen binding. However, with site-specific biotinylation chemistry, the biotin linker is placed at the thiol bridge position, ensuring that antibody-antigen binding is not affected. This approach enhances the sensitivity of bioassays like ELISA and Western Blot. |
