Biotin is a naturally occurring metabolic enzyme factor that exhibits activity through the action of specific protein-biotin ligases. The physiological functions of organisms are mainly controlled and regulated by intracellular proteins. The biotinylation reaction of protein can be carried out by enzymatic biotinylation and chemical biotinylation. Enzymatic biotinylation is a biotinylation reaction facilitated by biotin protein ligase (BPL). Chemical biotinylation is usually the modification of amino groups of proteins with biotin-N-hydroxysuccinimide or similar acylating agents. Compared with chemical biotinylation, enzymatic biotinylation reaction has higher binding capacity and higher specificity, but the amount of protein that can be reacted is less. Biotin is a soluble small molecule produced by plants or prokaryotes, which can be non-covalently combined with avidin or streptavidin, making the biotin-avidin system widely used in the field of biotechnology. Avi tags are short peptide tags consisting of 15 amino acid residues that can be biotinylated by biotin ligase (BirA enzyme) in vivo or in vitro.
Chemical biotinylation is usually the modification of amino groups of proteins with biotin-N-hydroxysuccinimide or similar acylating agents. The chemical biotin is an effective tool for labeling proteins and biomacromolecules, reducing the requirement for specific binding and being more reactive.
