Biotin is a naturally occurring metabolic enzyme factor that exhibits activity through the action of specific protein-biotin ligases. The physiological functions of organisms are mainly controlled and regulated by intracellular proteins. The biotinylation reaction of protein can be carried out by enzymatic biotinylation and chemical biotinylation. Enzymatic biotinylation is a biotinylation reaction facilitated by biotin protein ligase (BPL). Chemical biotinylation is usually the modification of amino groups of proteins with biotin-N-hydroxysuccinimide or similar acylating agents. Compared with chemical biotinylation, enzymatic biotinylation reaction has higher binding capacity and higher specificity, but the amount of protein that can be reacted is less.
