Biotin, an essential micronutrient, is a small organic molecule that acts as a cofactor for biotin-dependent metabolic enzymes. Biotin can bind avidin or streptavidin with a very high affinity to form a biotin-avidin/streptavidin system. The high affinity between biotin and avidin or streptavidin has been widely used in biochemistry and molecular biology, usually for efficient protein detection or capture. Proteins can be covalently labeled with biotin chemically or enzymatically, which is one of the efficient protein labeling methods. The expressed biotin constructs can be fully biotinylated by co-expression with a plasmid overproducing E. coli biotin ligase. The degree of biotinylation in vivo can be promoted by adding ATP. Escherichia coli biotin ligase (BirA), a bifunctional protein that acts as a transcriptional repressor to regulate biotin biosynthesis genes, covalently attaches biotin to the side chain of lysine.
