Gene probes can be produced in a variety of ways and fall into three types: gene-specific probes, oligonucleotide probes, and polymorphism probes. Gene-specific probes are produced from specific mRNA by reverse transcriptase, which synthesizes a complementary DNA copy (cDNA) from the mRNA. Traditional gene probes are synthesized from radioactive phosphate bases. Gene probes can be used in dot blot hybridization, where serially diluted DNA samples are preserved on DNA-binding membranes and complementary radiolabeled probes are in vitro, so that the amount of radioactive signal is directly proportional to the amount of target DNA present.
