The main method of biotin-avidin labeling technology

  • Bridging avidin-biotin labeling method (BAB): The bridging avidin-biotin labeling method can be divided into direct method and indirect method. Directly use free avidin or streptavidin as a bridging agent to combine biotinylated antibody complexes with labeled biotin, so that efficient detection can be performed. In the indirect method, after the antigen is combined with the specific antibody, the biotinylated second antibody is combined with the antigen-antibody complex, which improves the sensitivity of the reaction.
  • Avidin-biotin-peroxidase complex method (ABC): Mix avidin/streptavidin and enzyme-labeled biotin in a certain ratio to form a streptavidin-biotin-peroxidase complex.
  • Labeled avidin-biotin method (LAB): This method includes direct method and indirect method. The direct method is to directly link the labeled avidin or streptavidin to the biotinylated antibody in the immune complex for enzymatic color reaction. The indirect method uses a biotinylated secondary antibody to bind to the antigen, and its detection sensitivity is higher than that of the direct method.
  • Labeled streptavidin-biotin method (LSAB): The labeled streptavidin-biotin method is to detect cells and tissues by linking a biotin-labeled secondary antibody to streptavidin conjugated with horseradish peroxidase (HPR) or alkaline phosphatase (AKP).
  • Photobiotin method (Photobiotin): Photosensitive biotin is a chemically synthesized biotin derivative. The molecule contains a light-activated azide-nitrophenyl group. Under short-term irradiation of visible light, it can react with the basic group of nucleic acid to produce a biological Primer-labeled nucleic acid probes.
