Biotinylation refers to the process of covalently attaching biotin to proteins, nucleic acids or other biomolecules. Biotinylation reactions are fast, efficient and less susceptible to interference from environmental factors (such as temperature, pH, and proteases). Amine groups, mainly primary amine groups, are common reactive functional groups and are widely present in lysine side chains on the surface of proteins and in natural polymer materials. Amine groups can covalently synthesize amide bonds with functional groups of carboxylic acids, which are the most common biomarker targets and are usually used for biotinylation. Aminobiotinylation reagents are divided into common N-hydroxysulfosuccinimide (NHS) and sulfonated N-hydroxysulfosuccinimide (Sulfo-NHS). Common amination reagents are divided into ordinary N-hydroxysulfosuccinimide (NHS) and sulfonated N-hydroxysulfosuccinimide (Sulfo-NHS). These aminobiotinylation reagents are highly hydrophobic and permeable through cell membranes, enabling intracellular and extracellular targeting. The sulfonated N-hydroxysulfosuccinimide-activated biotin labeling reagent has better water solubility and is used in the labeling reaction.

The interaction between matterin-avidin/streptavidin is the strongest interaction known so far and is not easily affected by environmental factors such as pH, temperature and organic solvents.
